Gentarget provides different sets of premade lentivirial particles for human or mouse iPS genes (see iPS particle product page). This premade lentiviral particles is the single vector method for iPS cell generation. Four mouse iPS factor, c-Myc, Klf4, Oct4 and Sox2 (MKOS) were cloned under the same Tetracycline inducible CMV promoter. All 4 factors were cloned in frame but separated by F2A signal fragment, which result in the expression of four individual factor via "ribosomal skipping" mechanism. In addition, the particles contains a RFP-blasticidin dual selection marker under RSV promoter, which provides a convenience mothod to monitor the iPS factor expression efficiency, and to sort the positve ips expression cells. The four iPS factor are expressed under an optional inducible CMV promoter. The premade particle can be used as constitutive expression of four iPS factors, but optionally, it can be used as an tetracycline inducible expression of iPS factors. To used as inducible expression, the repressor protein (TetR) must be present inadvance to stop the expression, then the expression of iPS factors only occurs when the Tetracycline or Dox was added into the cell culture. This particle was produced by co-transfection of the chained iPS inducible lentivector with a packaging mix (Cat# HT-pack) into a 293T packaging cells (cat# TLV-C).
