Utilizing our engineered lentivectors and the packaging system, Gentarget produced Ready-To-Use lentiviral particles (LP) for 4 common used fluorescent proteins, GFP, RFP, CFG and YFP. Each fluorescent protein was codon optimized to demonstrated strongest fluorescent signals, and expressed under three different promoter. The proprietary suCMV promoter demonstrated the highest expression levels in majority cell types. The engineered EF1a promoter is non-tissue specific, highly expressed in all cell types, and less likely be silenced after long-term culture. The CAG promoter ( a hybrid of the CMV enhancer and chicken beta-actin promoter) has the highest expression level in embryonic stem cells. Please see the schematic lentivector structure below. It has different antibiotic marker, allowing selection of transduced stable cells.
The particles were pseudotyped with VSV-G protein and packed in 293T cell. Each particle was provided as 200 μL/vial containing >1 x 107-9 IFU. (Infection Function Unit) /ml.
They are packed in two formats:
- Particles are provied in DMEM medium with 10% FBS and 60ug/ml polybrene.
- Particles are concentrated, purified and buufer exchanged into PBS as In Vivo ready status.
The pre-made control lentiviruses for 4 fluorescent proteins are ideal for testing the transduction efficiency for the cells of your interest, cell tracking, stable cell line construction and enzymatic assays both in vivo or in vitro. They are extremely easy to use, no need for any mixing or addition, simply add certain amount of virus into your cultured cells. You will be able to observe the expression in 72 hours.
