Control lentivirus for target over expression and Knockdown shRNA:
GenTarget provides pre-made negative control lentiviruses for all of our lentiviral expression systems; these include controls for target overexpression, shRNA expression, and optional inducible expression lentiviruses.
1. Target over expression control lentivirus:
The negative control virus for target overexpression lentivirus contains a Null spacer sequence at the target position under either an optional inducible CMV promoter (TetCMV) or an enhanced EF1a promoter. Due to its lack of a start codon, the Null sequence allows packaging of the lentivirus but not translation into a protein. See control vector scheme below. The control virus serves as the negative control of lentivurs treatment for the same lentivector backbone. It validates the specificity of any target expression at the testing cells.
The negative expression control lentiviruses contain an antibiotic selection marker corresponding to each target expression lentivector. Please select the control lentivirus with the same promoter and antibiotic marker as your target expression lentivirus. See each control virus product link for details.
2. shRNA expression control lentivirus:
The negative control shRNA lentivirus contains a negative control shRNA sequence under either the optional inducible H1 promoter or constitutive U6 promoter. The Neg-control shRNA lentivirus controls for non-specific knockdown level; it validates the off-target effects of shRNA treatment. See control vector map scheme below..
The negative expression control lentiviruses contain an antibiotic selection marker corresponding to each shRNA expression lentivector. Please select the control lentivirus with the same promoter and antibiotic marker as your shRNA expression lentivirus. See each shRNA control virus product link for details.
3. Optional inducible expression control lentivirus:
GenTarget provides pre-made control viruses for optional inducible expression , used for controls in the a specific human, mouse or rat’s gene over-expression lentivirus products, or used to validate the optional inducible system in your specific cell lines. (See vector scheme above).
The inducible controls contain GFP or RFP under control of the inducible promoter, enabling very convenient verification of tetracycline inducible effects by simple monitoring of GFP or RFP signal under a microscope. The controls also contain a fluorescent-antibiotic fusion dual marker under a RSV promoter which is not affected by induction. The dual marker provides a convenient way to monitor viral transduction efficiency and select positive cells by either antibiotic killing or fluorescence sorting.
