Description
Pre-packaged rAAV in Serotype-DJ, constitutively express an Anti-CD19 CAR (Anti-CD19-ScFv-41BB-CD3ζ) with N-terminal 3X FLAG (DDK) tag, under a enhanced CMV Promoter (which is tested strong in T cells and B cells).
The ScFv of anti-CD19 (Click to see its sequence) (the variable region sequences of the FMC63 monoclonal antibody which recognizes human CD19), is fusion with the CAR (Chimeric Antigen Receptor) of “CD8 hinge, 4-1BB co-stimulatory domain, and CD3ζ signaling domains”. Its expression can be detected by anti-FLAG (DDK tag) antibody. The following scheme showed the AAV vector’s core structure.
CD19 is a marker of in most B cell leukemias and lymphomas but not in any normal tissue other than the B cell lineage, CD19 is used to diagnose cancers that arise from this type of cell, and is used as the target for CD19-targeted therapies. The AAV product is used as CAR-T targeted killing of the CD19 positive cancer cells. See Product Manual for details.

AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
