Pre-packaged rAAV in Serotype-DJ, constitutively express an Anti-human CD22 CAR [(VL-G4S-VH)-CD28-CD3ζ] with N-terminal 3X FLAG (DDK) tag, under a enhanced CMV Promoter (which is tested strong in T cells and B cells). See Product Manual for details.
The Anti-h CD22 ScFV (Click to see its sequence), is fusion with the CAR (Chimeric Antigen Receptor) of “CD8 hinge, CD28 co-stimulatory domain, and CD3ζ signaling domains”. Its expression can be detected by anti-FLAG (DDK tag) antibody and anti G4S (GGGGS) antibody . The following scheme showed the AAV vector’s core structure.
CD22 is a cell surface glycoprotein expressed on B cells. It is involved in B cell activation and regulation. CD22 is a target in CAR-T therapy for B-cell malignancies, especially in cases where CD19-targeted CAR-T cells may face challenges. CD22 CAR-T cells offer an alternative approach in certain B-cell lymphomas and leukemias. This CAR construct assembled the identified anti-human CD22 monoclonal antibodies’ coding sequence (clone m971, 704Y). (see Reference1, Reference2).

AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
