Description
Pre-packaged rAAV in Serotype-DJ, constitutively express an Anti-human HLA-A2 CAR [(VL-G4S-VH)-41BB-CD3ζ] with N-terminal 3X FLAG (DDK) tag, under a enhanced CMV Promoter (which is tested strong in T cells and B cells). See Product Manual for details.
The Anti-h HLA-A2 ScFV (Click to see its sequence), is fusion with the CAR (Chimeric Antigen Receptor) of “CD8 hinge, 4-1BB co-stimulatory domain, and CD3ζ signaling domains”. Its expression can be detected by anti-FLAG (DDK tag) antibody and anti G4S (GGGGS) antibody . The following scheme showed the AAV vector’s core structure.
HLA-A2 is a major histocompatibility complex (MHC) class I molecule involved in presenting antigens to cytotoxic T cells. HLA-A2 is not a traditional target for CAR-T therapy, as CARs typically recognize cell surface antigens rather than MHC molecules. CARs anti HLA is used to engineering human regulatory T cells (Tregs), and is a promising strategy to induce immune tolerance in transplants. This CAR construct assembled the coding sequence of the human monoclonal anti-HLA-A2 antibody (clone BB7.2 ), recognize HLA-A*02:01. (see Reference1, Reference2).

AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
