Description
Pre-packaged rAAV in Serotype-DJ, constitutively express an Anti-human TGFβ CAR [(VL-G4S-VH)-CD28-CD3ζ] with N-terminal 3X FLAG (DDK) tag, under a enhanced CMV Promoter (which is tested strong in T cells and B cells). See Product Manual for details.
The Anti-h TGFβ ScFV (Click to see its sequence), is fusion with the CAR (Chimeric Antigen Receptor) of “CD8 hinge, CD28 co-stimulatory domain, and CD3ζ signaling domains”. Its expression can be detected by anti-FLAG (DDK tag) antibody and anti G4S (GGGGS) antibody . The following scheme showed the AAV vector’s core structure.
TGF-β is often associated with immunosuppressive effects in the tumor microenvironment. High levels of TGF-β can contribute to immune evasion by inhibiting the activity of cytotoxic T cells and promoting the generation of regulatory T cells (Tregs). To counteract the immunosuppressive effects of TGF-β in the tumor microenvironment, the CAR-T cells are developed to resist TGF-β-mediated suppression. This CAR construct assembled the coding sequence from some identified anti-human TGFβ antibodies (see Reference).

AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
