Pre-packaged rAAV in Serotype-DJ, constitutively express the human Stem Cell factor, KLF4 (NM_004235, gene_synonym=EZF; GKLF) gene, under a modified CMV Promoter, No-tag. The following scheme showed the AAV vector’s core structure. See Product Manual for details.
Induced Pluripotent Stem Cells (iPSCs) can be generated with a set of transcription factors or stem cell factors. AAV expressed stem cell factors generate “footprint-free” iPSCs with no permanent genetic alterations, which may be more favorable due to the non-integrating nature and better safety profile.
AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle. To determine the serotype that best suits your desired cell types, you can use the AAV-GFP Serotype Screening Kit.
