Description
Pre-packaged rAAV in Serotype-DJ, constitutively express human TERT gene (NM_198253, gene_synonym=CMM9; DKCA2; DKCB4; EST2; hEST2; hTRT; PFBMFT1; TCS1; TP2; TRT), under a enhanced EF1a Promoter. The EF1α promoter is a better choice than CMV in cell types where CMV activity is low or where promoter silencing is a concern, as EF1α is significantly less prone to silencing.

Expression of Human telomerase reverse transcriptase (hTERT) in primary human cells, prevent telomere shortening, results in cell immortalization. hTERT is Widely used to immortalize fibroblasts, epithelial cells, and stem cells for long-term culture. AAV generally does not integrate into the host genome, thus AAV generates Footprint-Free Immortalization (extend the lifespan of cells without permanent genetic modification). AAV does not have the risk of insertional mutagenesis—a major safety concern when use Lentivirus. Cell immortalization requires sustained hTERT expression. AAV-mediated immortalization is not permanent and persists only as long as AAV-driven hTERT expression is maintained.
AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
