Expression of Large-T-antigen of simian virus (SV40-LT) in primary cells, is a well-established tool for cell immortalization. However, SV40-LT disrupts tumor suppressors, increasing the risk of transformation or chromosomal aberrations. AAV generally does not integrate into the host genome, thus AAV generates Footprint-Free Immortalization (extend the lifespan of cells without permanent genetic modification). AAV does not have the risk of insertional mutagenesis—a major safety concern when use Lentivirus. Cell immortalization requires sustained SV40-LT expression. AAV-mediated immortalization is not permanent and persists only as long as AAV-driven SV40-LT expression is maintained.
AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
