Pre-packaged rAAV in Serotype-DJ, constitutively express MART-1 specific T Cell Receptor (TCR) (Clone DMF5), under a enhanced CMV Promoter (which is tested strong in T cells and B cells). See Product Manual for details.
This TCR construct (Click to see its sequence) specifically recognizes a Peptide (peptide 2: ELAGIGILTV ) in antigen MART-1 (Melanoma-associated antigen recognized by T cells-1, or Melan-A). The TCR α chain variable region with a transmembrane (TM) domain is linked with TCR β chain variable region with a transmembrane (TM) domain, by F2A element, as “TCRα(V)/Tm-F2A-TCRβ(V)/Tm”. MART-1, also known as Melan-A, is a protein antigen found on the surface of melanocytes. The MART-1 antigen is a target in immunotherapy for patients with melanoma. TCRs targeting MART-1 recognize the antigen on melanoma cells, leading to T cell activation and cytotoxic T cell response to malignant melanoma cells. References: Ref 1, Ref 2.

AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
