Pre-packaged rAAV in Serotype-DJ, constitutively express NY-ESO-1-specific T Cell Receptor (TCR) (Clone 1G4), under a enhanced CMV Promoter (which is tested strong in T cells and B cells). See Product Manual for details.
This TCR construct (Click to see its sequence, from Clone 1G4) express the antibody specifically recognizes the tumor epitope SLLMWITQC at NY-ESO-1 (157-165), aims to provide a targeted and effective treatment for certain types of cancers. References: Ref 1.
The TCR α chain variable region with a transmembrane (TM) domain is linked with TCR β chain variable region with a transmembrane (TM) domain, by F2A element, as “TCRα(V)/Tm-F2A-TCRβ(V)/Tm”. The following scheme showed the AAV vector’s core structure.

AAV Serotype-DJ is a synthetic serotype made from different serotypes using DNA shuffling. AAV-DJ serotype has the enhanced transduction efficiency in vitro and in vivo compared to wild-type serotypes with Broad Cell Tropism, great transduction rate in CNS, Liver, Kidney, Fibroblasts, muscle.
