Description

Premade Nano-Lantern Lentivirus for imaging of dCas9 enzyme in Nuclear area. It allows for the tracing of dcas9 in targeted locus in real-time in live cells and tissues. 

About dCas9 Enzyme:

CRISPR-Cas9 genomic gene-editing technology utilizes a guide RNA molecule (gRNA) to direct a Cas endonuclease to the genomic DNA targeted site. Then, the Cas enzyme creates a double-strand break at genome. spCas9 (from Streptococcus pyogenes ) is the most frequently used cas endonuclease in CRISPR.  The dCas9 (deficient Cas9 or Dead cas9) is the mutant of spCas9. It lacks the cleavage ability (the mutation removed endonuclease activity), but can still bind to any guide RNAs (gRNA), thus binding to genomic DNA targeted site by gRNA.

Nano-Lantern Cas9 probe: 

This Lentivirus construct the fusion of Orange-Nano-Lantern with the dCas9. It contains a Nuclear localization sequence, which allows real-time imaging of dCas9 at the targeted locus in Nuclear area. Upon gRNA added that target at a specific gene (which need to be made separately), the labeled dcas9 will be guided to the specific locus where the Luminescence signal can be detected in real-time.  Click Product Manual for product details.
