Description

Concentrated Lentivirus bicistronically expresses two individual proteins: nuclear permeable CRE recombinase and the RFP marker under the same suCMV promoter. CRE contains a NLS (nuclear localization signal) at its N-term. This nuclear permeant CRE increases the recombination events in vivo. A Puromycin marker was expressed under RSV promoter. Particles allows to sort or select transduced cells for long term expression via RFP signal or via Puromycin antibiotic selection.

NLS-CRE and RFP are constitutively expressed in high level, and optionally, also can be used as tetracycline-inducible expression as desired. RFP signal provides convenient, realtime monitoring the performance of the lentivirus.

Cre protein, from bacteriophage P1, catalyzes recombination between 34 base pair target sequences, named lox sites. CRE lentiviral particles can catalyze the jointing event In Vivo in the presence of lox sites. Purified CRE enzyme can join individual plasmids containing lox sites.

Particles was concentrated and resuspended in PBS solution, best suitable for hard-to-transduced cell line, or serum-sensitive cell lines and for in vivo application.
