Lentivirus express MART-1 specific (peptide 2: ELAGIGILTV ) T Cell Receptor (TCR), containing GFPPuromycin dual selection

The TCR construct express (under the enhanced EF1a promoter), the antibody specifically recognizes the Peptide 1 in antigen MART-1 (Melanoma-associated antigen recognized by T cells-1, or Melan-A). The TCR α chain variable region with a transmembrane (TM) domain  and β chain variable region with a transmembrane (TM) domain, are linked by F2A element, as “TCRα(V)/Tm-F2A-TCRβ(V)/Tm”. It also transduces a GFPPuromycin (fluorescent-antibiotic fusion ) dual selection gene under the separate RSV promoter, which allow to enrich or select the transduced cells via GFP sorting or Puromycin killing. see the Lentivector scheme below

About MART-1 (Melanoma Antigen Recognized by T cells 1):

MART-1, also known as Melan-A, is a protein antigen found on the surface of melanocytes. The MART-1 antigen is a target in immunotherapy for patients with melanoma. TCRs targeting MART-1 recognize the antigen on melanoma cells, leading to T cell activation and cytotoxic T cell response to malignant melanoma cells. References: Ref 1, Ref 2.
